The interest for the understanding of the immune mechanisms in the oyster, Crassotrea gigas has been considerably increased these last years, mainly because of economie losses due to diseases which affected the continuity of this species. The studies of these mechanisms contributes to a better understanding of the differential susceptibilities observed in this species against various pathogens. In the present study, our  goal was to define the ontogenesis of immune system in C. gigas during the development, by functional genomic approaches. The French ostreiculture history highlights the fragility of its production caused by stocks overexploitation and diseases. Particularly, the flat oyster production decreased a lot after the appearance of two parasitic diseases: marteilliose and bonamiose. The bonamiose is due to a parasite Bonamia ostreae. The study of interactions between the parasite and oysters hemocytes with flow cytometry allowed to identify modifications of hemocytes activities wh en active parasite is present. On the other hand, inactive parasite does not induce modification for the same hemocyte's activities. Since 1997, a new pathology seasonally occurs in new caledonian shrimp farms during the warm season and was named Summer Syndrome. Diseased Litopenaeus stylirostris shrimp suffer from a septicemic vibriosis which was attributed to V. nigripulchritudo. Preliminary studies based on a collection of V. nigripulchritudo strains have brought to light different virulence levels according to experimental infections results; three virulence statuses were defined: highly (HP), moderately (MP) and non pathogenic (NP). Isolation and characterization of genes functionally involved in ovarian development of the giant tiger shrimp Penaeus monodon by suppression subtractive hybridization (SSH) Genet. Mol. Biol.

Suppression subtractive hybridization (SSH) libraries between cDNA in stages I (previtellogenic) and III (cortical rod) ovaries of the giant tiger shrimp (Penaeus monodon) were established. In all, 452 ESTs were unidirectionally sequenced. Sequence assembly generated 28 contigs and 201 singletons, 109 of which (48.0%) corresponding to known sequences previously deposited in GenBank. Several reproduction-related transcripts were identified. The full-length cDNA of anaphase promoting complex subunit 11 (PmAPC11; 600 bp with an ORF of 255 bp corresponding to a polypeptide of 84 amino acids) and selenoprotein M precursor (PmSePM; 904 bp with an ORF of 396 bp corresponding to a polypeptide of 131 amino acids) were characterized and reported for the first time...

Study of interactions between flat oyster, Ostrea edulis, and the parasite Bonamia ostreae using flow cytometry and suppression subtractrive hybridization (SSH) ArchiMer

Bonamiosis due to the intrahaemocytic protistan parasite Bonamia ostreae is a European endemic disease affecting flat oysters Ostrea edulis. Control of bonamiosis requires a good knowledge of the parasite life cycle including host-parasite interactions. In this study, we investigated interactions between parasites and haemocytes by flow cytometry and Suppression Subtractive Hybridization (SSH). In both approaches, analyses were performed after 2 hours of contact between haemocytes and parasites purified from highly infected flat oysters.
